Tetrazine-Dox

Tetrazine-Doxorubicin(Tz-Dox) aims to achieve tumor specific drug activation through bioorthogonal chemical reactions, thereby improving therapeutic efficacy and reducing systemic toxicity.

Tetrazine group: As a switch in orthogonal biological reactions, it can undergo a rapid, copper free Diels Alder (IEDDA) reaction with trans cyclooctene (TCO), triggering drug release. In Tz-Dox, tetrazine not only acts as a reaction site, but also as a masking group for prodrugs, inhibiting Dox activity until the target site is activated.
