Calcein AM(Calcein acetoxymethyl ester) is a fluorescent dye that can penetrate cells and is used to measure cell viability. It is also used as a green fluorescent probe for measuring the viability of eukaryotic cells or mitochondrial permeability transition pore (MPTP). In recent years, Calcein AM has been widely used in cell viability analysis.Calcein AM itself has no fluorescence. After entering the cell, it is hydrolyzed by endogenous esterase to form calcein, a polar molecule with strong negative charge and unable to penetrate the cell membrane, which is retained in the cell, and calcein can emit strong green fluorescence. Compared with other similar probes, Calcein AM is one of the most ideal fluorescent probes for staining living cells because of its low cytotoxicity, almost no effect on cell function, such as cell proliferation or lymphocyte chemotaxis, and low sensitivity to pH.
