Based on the aggregation induced luminescence material tpe-tpp,the characteristics of photon fluorescence were studied.Using tpe-tpp as a fluorescence probe,the advantages and disadvantages of three photon and two-photon fluorescence imaging technology under 1020nm and 810nm femtosecond laser excitation were compared.Several advantages of three photon fluorescence imaging technology based on tpe-tpp were proposed,including high photobleaching resistance,low light damage, high spatial resolution and high signal to background ratio.Three photon fluorescence imaging of tpe-tpp was also used to observe the change trend of mitochondrial membrane potential in real time and with high sensitivity.In addition,tpe-tpp can be used to label neurons and microglia in mouse brain.Through microinjection,the real-time and long-time two-photon fluorescence imaging of microglia in mouse brain excited by 740nm femtosecond laser was realized.Three photon fluorescence imaging of brain tissue sections is performed to obtain high-quality imaging, which has great significance and application prospects for the study of deep nervous system activity in vivo. |
